牛源金黄色葡萄球菌α溶血素单链抗体对小鼠乳腺的保护作用

中文摘要:为探究治疗奶牛乳腺炎的抗生素替代性生物制剂,研究将前期获得的特异性针对金黄色葡萄球菌毒力因子α溶血素的单链抗体scFvZZ107,在小鼠感染金黄色葡萄球菌乳腺炎模型中验证其保护作用。结果显示,3种不同剂量(5、10、15 μg)单链抗体处理组乳腺组织内细菌数量分别为25.42±1.3、3.33±0.53和4.8±0.78 lgcfu/mg,IL-1α的含量分别为49±7.91、31.37±2.26和57.34±4.48 pg/mL,IL-1β含量分别为40.91±7.39、47.81±4.78和39.2±2.28 pg/mL,IL-8的含量分别为48.64±1.86、41.65±3.07和45.74±1.56 pg/mL,与生理盐水处理组相比,单链抗体中、高剂量处理组乳腺内金黄色葡萄球菌检出量显著减少(P<0.05),且IL-1α、IL-1β与IL-8的含量均显著降低(P<0.05)。通过观察小鼠乳腺组织HE和TUNEL染色情况,与生理盐水处理组相比,scFv中、高剂量处理组乳腺组织更加完整,凋亡细胞的数量也更少,表明scFvZZ107对小鼠乳腺具有良好的保护作用。

Protective effect of single-chain antibody against α hemolysin of Staphylococcus aureus on mouse mastitis
Abstract:In order to explore antibiotic-alternative biological agents for the treatment of mastitis in dairy cows,in this study,a previously-obtained single-chain antibody scFvZZ107 specific to S.aureus virulence factor alpha hemolysin was administered in a mouse model of S.aureus infection mastitis to confirm its protection.As a result,the numbers of bacteria in the mammary glands of the three treatment groups with 5,10 and 15 μg of scFvZZ107 were 25.42±1.3,3.33±0.53 and 4.8±0.78 lgcfu/mg,respectively.The contents of IL-1α were 49±7.91,31.37±2.26 and 57.34±4.48 pg/mL,respectively.The contents of IL-1β were 40.91±7.39,47.81±4.78 and 39.2±2.28 pg/mL,respectively.The contents of IL-8 were 48.64±1.86,41.65±3.07 and 45.74±1.56 pg/mL,respectively.In comparison with the saline-treated group,the numbers of S.aureus in the mammary glands treated by moderate or high doses of scFv were significantly decreased (P<0.05),and the contents of IL-1α,IL-1β and IL-8 were lower (P<0.05).The HE and TUNEL staining revealed that the proportions of intact mammary gland tissues were higher and the numbers of apoptotic cells were lower than that of the saline-treated group.Collectively,our present findings indicated that scFvZZ107 had a protective effect on mouse mastitis.
